What AI Technology Is Doing for Longevity Now

In March 2023, MIT Technology Review revealed that Sam Altman, the CEO of OpenAI (ChatGPT), was the mystery investor behind the $180 million investment into stealth startup Retro Biosciences, a biotech company with the ambition of “adding 10 years to the human lifespan.” This investment marks the latest tech entrepreneur expressing their interest in longevity science and a new connection with innovative AI technology.

According to February 2023 reports, AI is continuing to gain traction in healthcare applications. Currently, the market is estimated at $14.6 billion (USD) with a compound annual growth rate (CAGR) of 47.6%, with solutions spread across various healthcare fields, such as patient data and risk analysis, precision medicine, cybersecurity, lifestyle management, and drug discovery.

The increasing convergence of AI technology and longevity science is sparking advancements in the sector, with established businesses, start-ups, and researchers utilizing the technology. Most recently, scientists explored how ChatGPT, an AI-based language model, was able to predict Alzheimer’s in 80% of cases when analyzing speech. However, it is not the only implementation.

Recent clinical trials with stem cells to slow or reverse normal aging processes

Stem cells (SCs) are undifferentiated cells which can proliferate indefinitely or differentiate into progenitor cells and end-phase differentiated cells (becoming pluripotent) (Mayo, 2021; Slack, 2022). Human embryonic SCs (hE-SCs) are found in the inner cell mass of the blastocyst; h E-SC research raises ethical concerns (Lo and Parham, 2009), and h E-SC transplantation in vivo can lead to the formation of large tumors called teratomas (Blum and Benvenisty, 2008).

Small numbers of adult SCs are found in some organ “niches”, including the bone marrow, where hematopoietic progenitor cells (HPC) replenish blood and immune cells. In 1958, Mathe et al. (1959) successfully performed the first adult SC therapy on five workers who had received high-dose accidental irradiation at the Vinca Nuclear Institute in Yugoslavia. After transfusions and grafts of homologous adult bone marrow, all workers survived (Mathe et al., 1959).

For years, the human umbilical cord was a waste material and, unlike h E-SCs, its use does not raise ethical concerns. In 1988, Gluckman et al. (1989) successfully performed the first human cord blood transplant in a child with Fanconi’s anemia. Since then, numerous public and private cord blood banks have been established worldwide for the cryopreservation of cord blood in view of its transplantation (Gluckman, 2011).

Genomes from 240 mammalian species reveal what makes the human genome unique

Over the past 100 million years, mammals have adapted to nearly every environment on Earth. Scientists with the Zoonomia Project have been cataloging the diversity in mammalian genomes by comparing DNA sequences from 240 species that exist today, from the aardvark and the African savanna elephant to the yellow-spotted rock hyrax and the zebu.

This week, in several papers in a special issue of Science, the Zoonomia team has demonstrated how can not only shed light on how certain species achieve extraordinary feats, but also help scientists better understand the parts of our genome that are functional and how they might influence health and disease.

In the new studies, the researchers identified regions of the genomes, sometimes just single letters of DNA, that are most conserved, or unchanged, across mammalian species and millions of years of evolution—regions that are likely biologically important. They also found part of the genetic basis for uncommon mammalian traits such as the ability to hibernate or sniff out faint scents from miles away. And they pinpointed species that may be particularly susceptible to extinction, as well as genetic variants that are more likely to play causal roles in rare and common human diseases.

DNA methylation markers for increased risk of schizophrenia identified for first time in newborns

An international research team led by investigators at Virginia Commonwealth University has identified for the first time markers that may indicate early in life if a person has susceptibility to schizophrenia.

The ability to predict the risk of developing later in life may allow early detection and intervention, which the researchers hope can reduce the impact of the disease on individuals, families and communities. Their results have been published in Molecular Psychiatry.

Schizophrenia is a serious psychiatric disorder that is most often detected in young adulthood. It affects as much as 1% of the and can cause debilitating effects such as a sense of losing touch with reality. People with the disorder are up to three times more likely to die early and often face discrimination, social isolation and debilitating physical illness, according to the World Health Organization.

New Therapy Found to Prevent Aggressive Brain Cancer Recurrence in Mice

A new gel-based treatment for glioblastoma—a highly aggressive form of brain cancer—has shown to be 100% effective at preventing recurrence in mice. Researchers hope the therapy will translate well into human physiology, where it could help resolve tens of thousands of cancer diagnoses every year.

Glioblastoma manifests as a tumor growing on the brain or spinal cord. While many glioblastoma patients have the tumor surgically removed, the mass often returns, even in cases involving post-surgical radiation or chemotherapy. The disease is so persistent that the average patient lives only 12 to 16 months after diagnosis, making glioblastoma one of the most lethal forms of cancer currently understood.

High rate of concurrent retinal non perfusion encountered in Pediatric patients with optic nerve hypoplasia

A new study published in the American Journal of Ophthalmology suggests that there seems to be a significant rate of concomitant retinal nonperfusion in pediatric optic nerve hypoplasia (ONH) patients.

This study was carried out by Natasha da Cruz and colleagues to report the correlation between peripheral retinal nonperfusion, secondary problems, and optic nerve hypoplasia in pediatric patients.

The Bascom Palmer Eye Institute conducted the Retrospective case series investigation between January 2015 and January 2022. Age under 18 years old, a clinical diagnosis of optic disc hypoplasia, and an acceptable-quality FA were the inclusion criteria.

Study suggests that maintaining normal vitamin D levels may benefit patients with advanced skin cancer

New research indicates that for patients with advanced skin cancer, it may be important to maintain normal vitamin D levels when receiving immunotherapy medications called immune checkpoint inhibitors. The findings are published by Wiley online in CANCER, a peer-reviewed journal of the American Cancer Society.

Vitamin D has many effects on the body, including regulation of the immune system. To see whether levels of vitamin D might impact the effectiveness of immune checkpoint inhibitors, investigators analyzed the blood of 200 patients with advanced melanoma both before and every 12 weeks during immunotherapy treatment.

A favorable response rate to immune checkpoint inhibitors was observed in 56.0% of patients in the group with normal baseline vitamin D levels or normal levels obtained with vitamin D supplementation, compared with 36.2% in the group with low vitamin D levels without supplementation. Progression‐free survival—the time from treatment initiation until —in these groups was 11.25 and 5.75 months, respectively.

Researchers discover a novel genetic disorder associated with neurodevelopmental differences

Researchers from Children’s Hospital of Philadelphia (CHOP) and Princeton University have discovered a novel genetic disorder associated with neurodevelopmental differences. The discovery identified the disorder in 21 families from all over the world. The study “Abrogation of MAP4K4 protein function causes congenital anomalies in humans and zebrafish” is published in Science Advances today, April 26.

The as-yet unnamed disorder is the result of a series of rare variants in the MAP4K4 gene, which is involved in many signaling pathways, including the RAS pathway that normal cell growth, and is being investigated as druggable target for multiple disorders.

The researchers had documented several patients with craniofacial and neurodevelopmental issues that indicated a then-unknown genetic cause. They put out an international call for patients who seemed to fit these specific criteria. Ultimately, they were able to identify patients from 36 countries to determine whether there was a genetic linking them to their clinical issues.
